I got my Tables in Wonderland card at Guest Services in the first park I hit, Epcot on the outside of the park. You can get them both inside and outside at Guest Services at each park and I believe DTD but not at the hotel. Make sure TiW uses your AP when you register so you pay $75 instead of the FL resident and DVC rate of $100.
Lobby Concierge at the Disney Resorts sell Annual Passes but not renewals. The Lobby Concierge can only put the AP on a plastic Key to the World card, not on a paper ticket.

We were able to renew our TIW at the hotel concierge when we were there this past January. Annual passes I had to renew at home and I had to turn the voucher in at the park guest services window.
Right now Annual Passholders get a 10% discount at just about every merchandise location. Premium APs get 20% discount.
However all of the current AP perks are set to expire on May 31, 2012 and there is no information on whether or not a similar discounts will be offered beyond June 1.
